Ultrastructural Localization of Antigenic Sites in Paragonimus iloktsuenensis


Abstract
An immunoelectron microscopic observation of various antigenic sites of the lung fluke, adult Paragonimus iloktsuenensis, was performed by applying the indirect immunoperoxidase technique to localize the selective antigenic sites. The fixative
was
mixture of 1% paraformaldehyde and 0.2% glutaraldehyde in 0.1M phosphate buffer, pH 7.4. LR-white resin(EMS, hard grade), the embedding media, was polymerized at -20¡Éunder the ultraviolet light(365nm long-wave length) for 48 hours. Nickel
grids(EMS,
200 mesh) were coated with 2% collodion(in isoamyl-acetate) and 0.5% polyvinyl formvar. The antiserum produced from the rat infected with Paragonimus iloktsuenensis(10 weeks after infection) was used as a primary antibody. The secondary antibody
was
gold conjugated goat anti-rat IgG(particle size=10nm, Sigma).
The intestinal epithelial cells containing numerous antigenic granules which seem to be secreted into the intestinal lumen. The secretory granules and luminal contents were predominantly labeled by gold particles. The excretory bladder was also
proved
to be a highly antigenic organ, since its lamellated structures and contents were labeled by numerous gold particles. The antigenicities of secretory materials of the vitelline glands, testes, and ovaries were moderately recognizible, too. But
the
tegument was very weakly labeled by gold particles.
The above results suggests that the secretory substances from the intestinal epithelial cells and excretory contents from excretory bladder in the adult Paragonimus iloktsuenensis are the most potent antigenic stimulants to the host immune
system.
